Biology Seminar Series: Host-pathogen interactions during Coxiella burnetii infection

Join the Biology Department for its weekly Lester Newman Seminar Series. This week, hear from Madur Sachan, a Ph.D. candidate in the Raghavan Lab. His talk is titled, "Host-pathogen interactions during Coxiella burnetii infection." Plan to join the seminar by 11:50 a.m.

Abstract: When infecting a human macrophage, Coxiella burnetii, the causative agent of Q fever, has the unique ability to replicate within a lysosome-derived vacuole (CCV). Previous studies have shown that inhibition of apoptosis — a programmed cell death, is a critical component of C. burnetii’s pathogenicity. However, nothing is known about the roles host microRNAs (miRNAs), an important class of non-coding regulatory RNAs, potentially play in this process. We investigated the functions of miRNAs during Coxiella infection and found that several miRNAs, including miR-143-3p and miR-146a-5p, can contribute to the subversion of host cell apoptosis, intracellular trafficking, and cytokine signaling. Further, single-cell transcriptomics and miRNA-mRNA interaction analysis showed that miRNAs likely act as molecular switches in macrophage polarization and plasticity during infection, along with a potential role in nutritional immunity. Collectively, our data revealed multiple important roles for miRNAs during C. burnetii infection of human macrophages.
